Target and Hazard: African swine fever (ASF) is caused by African swine fever virus (ASFV) and is a highly lethal disease of pigs, listed as a Class I animal disease in China. Affected pigs show high fever, cyanosis, vomiting and bloody diarrhoea, with mortality up to 90–100% in acute cases. No safe and effective commercial vaccine is currently available, so control relies on early diagnosis, quarantine, stamping-out and biosecurity. Recovered and subclinically infected pigs may carry the virus for long periods, acting as hidden sources of infection and driving continued spread.
Significance of Testing: The kit detects ASFV-specific antibodies in serum or plasma for epidemiological investigation of ASF, screening of recovered and subclinically infected pigs, and outbreak trace-back. Importantly, antibody positivity reflects infection or a history of infection and does not indicate immune protection; this product is not intended for, and cannot be used for, evaluating vaccine efficacy. Antibody and antigen testing are complementary and together form an important part of ASF surveillance and eradication.
Principle: Blocking ELISA: microplates are coated with ASFV-specific recombinant antigen, and antibodies in the sample compete with a fixed amount of enzyme-labelled anti-ASFV monoclonal antibody for the antigenic sites; the higher the sample antibody level, the weaker the colour development (the higher the blocking rate).
Performance and Applications: 96-well format; total assay time approximately 2–2.5 hours; read at 450 nm and interpreted by blocking rate, giving objective results suited to batch testing; storage at 2–8 °C. For ASF epidemiological investigation and monitoring by large pig farms, animal disease prevention and control institutions and diagnostic laboratories. Method: blocking ELISA | Samples: serum, plasma (pig) | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm, interpreted by blocking rate | Format: 96-well plate | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods are subject to our internal QC report.
