Description

Target and Hazard: Brucellosis is a zoonosis caused by bacteria of the genus Brucella, affecting cattle, sheep, pigs and other livestock. It is a Class II animal disease in China and subject to statutory reporting. Infected animals mainly show abortion, stillbirth, retained placenta, infertility, orchitis and arthritis, causing severe reproductive failure and losses in livestock production. People can be infected through contact with abortus material, secretions or unpasteurised milk and develop undulant fever — an occupational and public-health risk. B. abortus, B. melitensis and B. suis must be distinguished epidemiologically.

Significance of Testing: The kit detects Brucella antibodies in serum for mass screening and eradication of brucellosis in cattle, sheep and pig herds: regular testing identifies antibody-positive animals for removal, gradually building brucellosis-free herds. It is also used for quarantine testing of incoming animals and epidemiological investigation, and is a core tool for antibody monitoring in brucellosis control and eradication programmes.

Principle: Indirect ELISA: microplates are coated with Brucella-specific antigen; antibodies in the sample bind the coated antigen, an enzyme-labelled secondary antibody is then bound and substrate development follows, with colour intensity related to the sample antibody level.

Performance and Applications: 96-well format; total assay time approximately 2–2.5 hours; read at 450 nm and interpreted by OD value, giving objective results suited to batch testing; storage at 2–8 °C. For mass screening and eradication monitoring by large cattle, sheep and pig farms, animal disease prevention and control institutions and third-party laboratories. Method: indirect ELISA | Samples: serum (cattle/sheep/pig) | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm, interpreted by OD value | Format: 96-well plate | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods such as the serum agglutination test (SAT) are subject to our internal QC report.
