Description

Target and Hazard: Porcine circovirus type 2 (PCV2) is the principal pathogen of porcine circovirus-associated disease, causing postweaning multisystemic wasting syndrome (PMWS), porcine dermatitis and nephropathy syndrome (PDNS) and reproductive failure. Pigs are the natural host, and the virus spreads by oronasal contact, through semen and across the placenta. Infected pigs grow slowly, waste, show a higher feed conversion ratio and increased mortality, and lymphoid damage causes immunosuppression that raises herd susceptibility to porcine reproductive and respiratory syndrome virus, Mycoplasma hyopneumoniae and other pathogens — a major source of hidden losses on intensive farms. Porcine circovirus disease is a Class III animal disease in China.

Significance of Testing: The kit detects PCV2-specific IgG antibody in serum to assess seroconversion rate and herd immune qualification after vaccination and to monitor infection pressure and infection history, supporting optimisation of vaccination programmes. Antigen or nucleic acid testing reflects current virus carriage, whereas antibody testing reflects immune response and past exposure; used together they give a complete picture of PCV dynamics in the herd.

Principle: Indirect ELISA: microplates are coated with recombinant PCV2 Cap protein antigen; PCV2 antibodies in the sample bind to it and are recognised by an enzyme-labelled anti-pig IgG secondary antibody. Substrate is added and colour intensity is directly proportional to the sample antibody level.

Performance and Applications: 96-well format; total assay time approximately 2–2.5 hours; read at 450 nm on a microplate reader for objective results suited to batch testing; storage at 2–8 °C. Intended for post-vaccination monitoring on pig farms, herd serosurveys by animal disease control institutions and third-party laboratories, and quarantine testing of incoming animals. Method: indirect ELISA (recombinant PCV2 Cap protein coated) | Samples: porcine serum/plasma | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm, interpreted from the S/P or OD value | Format: 96-well plate with positive and negative controls | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods are subject to our internal QC report.
