Target and Hazard: Avian influenza is caused by avian influenza virus (AIV), a type A influenza virus of the family Orthomyxoviridae, to which chickens, ducks, geese and wild birds are susceptible. Highly pathogenic strains among the H5 subtype (e.g. H5N1) can cause high morbidity and mortality in poultry, are classified as a Class I animal disease in China and can infect humans, giving them major public-health importance. The virus spreads via the respiratory and digestive routes and contaminated materials, and transmits extremely rapidly in intensive housing and live-bird markets; early detection and response are key to outbreak control.
Significance of Testing: This duplex card carries a universal test line and an H5-subtype test line on one strip. The universal line recognises the type-specific antigen shared by all type A influenza viruses (e.g. NP/M protein) for broad screening of type A AIV infection in a flock, while the H5 line further identifies the H5 subtype, achieving combined universal and subtype detection for preliminary on-site differentiation of H5 highly pathogenic avian influenza. It is intended for on-site investigation at poultry farms, live-bird markets, slaughterhouses and border quarantine. A positive result must, in accordance with national regulations, be reported and submitted to the laboratory for confirmation.
Principle: Colloidal gold immunochromatographic assay (double-antibody sandwich): the universal type A AIV antigen and the H5-subtype antigen in the sample each bind to their corresponding gold-labelled antibody, migrate along the nitrocellulose membrane and are captured by the specific antibody coated on the universal test line (T1) and the H5-subtype test line (T2), producing visible lines; the control line (C) confirms reagent and procedure validity.
Performance and Applications: No instrument required; visual reading within 10–15 minutes; samples: tracheal/cloacal swabs, tissue homogenate; stored and shipped at room temperature — suitable for rapid on-site investigation at poultry farms, live-bird markets, slaughterhouses, primary veterinary stations and border quarantine. Method: colloidal gold immunochromatographic assay (double-antibody sandwich) | Samples: tracheal/cloacal swabs, tissue homogenate | Time to result: 10–15 min | Reading: visual assessment of T1 (universal)/T2 (H5 subtype)/C lines | Format: tests/box (as stated on the label) | Storage: 2–30 °C, cool and dry; shelf life 24 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with RT-PCR/virus isolation are subject to our internal QC report.
