Target and Hazard: Classical swine fever (CSF, hog cholera) is caused by classical swine fever virus (CSFV, genus Pestivirus, family Flaviviridae) and is a highly contagious acute febrile disease, classified as a Class II animal disease in China. Affected pigs show persistent high fever, anorexia, conjunctivitis, skin petechiae and gastrointestinal signs, and mortality is high in the acute form; infection of pregnant sows causes transplacental transmission, leading to abortion, stillbirth, mummified fetuses and weak piglets, with persistent virus carriage and immunosuppression in the herd. Rapid spread and severe losses make CSF a priority target for control and eradication in the pig industry.
Significance of Testing: This kit uses a blocking format to detect CSFV-specific antibodies in serum. Because sample antibodies compete with a monoclonal antibody for the antigenic sites, the reaction is highly targeted and little affected by differences between virus strains and vaccine strains, making it well suited to neutralising-antibody and immune-efficacy assessment. It is used for immune-qualification monitoring, differentiation between vaccine-induced and field-virus antibodies, and antibody tracking during the establishment of disease-free areas and on-farm eradication, complementing antigen testing to cover both the infection and the immunity pathways.
Principle: Blocking ELISA: microplates are coated with CSFV-specific antigen; antibodies in the sample compete with a fixed amount of enzyme-labelled anti-CSFV monoclonal antibody for the antigenic sites. The higher the sample antibody level, the less labelled antibody binds and the weaker the colour (the higher the percent blocking), which is used to grade antibody levels.
Performance and Applications: 96-well format; total assay time approximately 2–2.5 hours; read at 450 nm and interpreted by percent blocking, giving objective results suited to batch testing; storage at 2–8 °C. Intended for vaccination monitoring on large-scale pig farms and for eradication and epidemiological programmes run by animal disease control institutions and third-party diagnostic laboratories. Method: blocking ELISA (CSFV-specific antigen coated, enzyme-labelled monoclonal antibody competitor) | Samples: porcine serum/plasma | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm, interpreted as percent blocking | Format: 96-well plate with positive and negative controls | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods are subject to our internal QC report.
