Target and Hazard: Pseudorabies is caused by pseudorabies virus (PRV). Pigs are the natural host and source of infection, while other domestic animals and wildlife usually develop fatal disease after infection. The virus spreads by oronasal contact, mating and across the placenta: infected pregnant sows abort or produce stillborn and mummified fetuses, piglets show neurological signs, vomiting and diarrhoea with high mortality, and growing pigs develop clear respiratory signs and reduced weight gain, directly causing reproductive losses and delayed marketing. Pseudorabies is a Class III animal disease in China, and commercial farms generally control it through vaccination combined with eradication measures.
Significance of Testing: Glycoprotein gB is the major immunogenic protein of PRV and is essential for virus replication; both vaccination and field infection induce gB antibodies, so this marker reflects the herd immune response. The kit is used to assess seroconversion rate and immune qualification after vaccination, to monitor how well the vaccination programme is being implemented and how maternally derived antibodies decline, providing a basis for adjusting vaccination timing and the number of doses. Used together with a gE antibody kit, it enables dual monitoring of immune qualification and field infection.
Principle: Blocking ELISA: microplates are coated with recombinant PRV gB protein; gB antibodies in the sample compete with a fixed amount of enzyme-labelled anti-gB monoclonal antibody for the antigenic sites. The higher the sample antibody level, the weaker the colour (the lower the S/N value), which is used for interpretation.
Performance and Applications: 96-well format; total assay time approximately 2–2.5 hours; read at 450 nm (630 nm reference) and interpreted by the S/N value for objective results; storage at 2–8 °C. Intended for vaccination-efficacy monitoring on commercial pig farms and for serological assessment by animal disease control institutions and third-party laboratories. Method: blocking ELISA (recombinant PRV gB protein coated, enzyme-labelled anti-gB monoclonal antibody competitor) | Samples: porcine serum/plasma | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm (630 nm reference), interpreted as the S/N value | Format: 96-well plate with positive and negative controls | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods are subject to our internal QC report.
