Target and Hazard: Pseudorabies virus (PRV) can infect pigs and many other domestic and wild animals, with pigs as the only natural host and reservoir. The virus can remain latent in the trigeminal ganglion for life and re-excrete under stress, regrouping or immunosuppression, creating a source of infection that is difficult to eliminate. Infected sows abort or produce stillborn and mummified fetuses, piglets develop neurological signs and die in large numbers, and growing pigs show clear respiratory signs with reduced weight gain. Pseudorabies is a Class III animal disease in China and one that breeding farms must control on a long-term basis.
Significance of Testing: Glycoprotein gE is the protein deliberately deleted from gene-deleted pseudorabies vaccines, and pigs vaccinated with a gE-deleted vaccine do not develop gE antibodies — so a gE antibody-positive serum result is direct evidence of field virus infection. This kit is therefore a core tool for PRV eradication programmes on breeding farms (building gE-antibody-negative herds): it monitors field infection rates, screens and removes gE-positive animals and verifies the effectiveness of vaccination and eradication measures. Used with gB antibody testing, it provides both the field infection status and the vaccine immune-qualification rate.
Principle: Blocking ELISA: microplates are coated with recombinant PRV gE protein; gE antibodies in the sample compete with a fixed amount of enzyme-labelled anti-gE monoclonal antibody for the gE antigenic sites, so the higher the sample antibody level the weaker the colour (the lower the S/N value), which is used to determine field infection status.
Performance and Applications: 96-well format with positive and negative controls; total assay time approximately 2–2.5 hours; read at OD630 nm and interpreted by the S/N value for objective, batch-scale testing; store at 2–8 °C, protected from light, shelf life 12 months. For field-infection monitoring and eradication assessment in breeding herds and for epidemiological investigation by animal disease control institutions and third-party laboratories. Method: blocking ELISA (recombinant PRV gE protein coated, enzyme-labelled anti-gE monoclonal antibody competitor) | Samples: porcine serum/plasma | Assay time: ~2–2.5 h | Reading: microplate reader at OD630 nm, interpreted as the S/N value (S/N > 0.70 negative, 0.60–0.70 suspect, ≤ 0.60 positive) | Format: 96-well plate with positive and negative controls | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with reference methods are subject to our internal QC report.
