Description

Target and Hazard: Foot-and-mouth disease (FMD) is caused by foot-and-mouth disease virus (FMDV, genus Aphthovirus, family Picornaviridae) and affects cloven-hoofed animals including pigs, cattle, sheep and goats. It is a Class I animal disease in China and a WOAH-listed disease requiring notification. Affected animals develop vesicles and erosions on the oral mucosa, hooves and teats, and young stock may die suddenly from myocarditis. Morbidity is high and spread extremely rapid; an outbreak requires stamping-out, movement control and trade restrictions, causing severe losses to livestock production and international trade.

Significance of Testing: Type O is one of the predominant circulating serotypes in China and neighbouring regions. This kit detects type O FMDV-specific antibodies in serum for post-vaccination antibody monitoring and immune-qualification assessment of inactivated vaccine programmes, supporting optimisation of vaccination schedules and evaluation of herd protection. Used together with non-structural protein (3ABC/NSP) antibody testing, it also allows differentiation between vaccinated and field-virus-infected animals — a key tool for FMD-free status and disease eradication.

Principle: Competitive ELISA: microplates are coated with type O FMDV-specific antigen, and antibodies in the sample compete with a fixed amount of enzyme-labelled monoclonal antibody for the antigenic sites. The higher the sample antibody level, the weaker the colour development (the higher the percent inhibition). The assay is suitable for bovine, ovine and porcine sera and avoids interference from species-specific secondary antibodies.

Performance and Applications: supplied in 96T, 192T and 480T configurations, with approximately 40–44 samples per plate including positive and negative controls; total assay time approximately 2–2.5 hours; read at 450 nm on a microplate reader and interpreted by percent inhibition (PI), giving objective results suited to batch testing; storage at 2–8 °C. For large-scale pig, cattle and sheep farms, animal disease prevention and control institutions and third-party testing laboratories. Method: competitive ELISA | Samples: serum (cattle/sheep/pig) | Assay time: ~2–2.5 h | Reading: microplate reader at 450 nm, interpreted as percent inhibition (PI) | Format: 96T / 192T / 480T | Storage: 2–8 °C, protected from light; shelf life 12 months | Note: sensitivity, specificity, limit of detection, intra-/inter-assay CV and agreement with the virus neutralisation test are subject to our internal QC report.
